Arabidopsis Seeds Sowing

Before lab

  1. Soil preparation

    1. Tray preparation: 12x4 flat tray with the rectangle tray underneath
    2. Prepare the soil: use Suremix, moist with water
    3. Fill the square little pots with soil, and put the pots into the 4x8 tray
    4. Bring the tray cover together with the filled trays to BSP
  2. Agarose preparation

    1. Make 0.1% low-melting agarose in water, which can be done in a microwave
    2. Add Tween 20 for a final concentration of 0.1% (100 μl/100 ml)

In lab seeds sowing procedure

  1. Label the flat with potted soil:
    • Mutant ID; Team ID; Date
  2. Obtain one tube of each of the following seeds:
    • WT Col-0, PLIP3-OX, EMS ID
  3. Label three Petri dishes with a marker: WT, PLIP3-OX, EMS ID
  4. Label the soil pots in the flat with the following arrangement:

  5. Use P1000 to take 2 ml of agarose into the petri dish
  6. Use P1000 to transfer the seeds from the tubes and add them to the 2 ml agarose in the petri dish, mixing well by gentle pipetting.
    Important: keep the seeds from different vials separated, DON’T mix the seeds.
  7. Use P200 (adjust to 100 μL) or P1000 (adjust to 200 μL) to pick the seeds up and drop each seed at the center of each quarter area of the pot. Each pot will have 4 seeds.
    Important:
    • Cut the smaller pipet tips (P200 tips) with a scissor if you use a P200. There is no need to cut the
      tip if you use P1000.
    • Change pipet tips when you start seeding from a new vial, this will avoid the cross contamination in case there are seeds stuck in the dropper or tip.
  8. Repeat the sowing until you finish all the pots. Add more agarose to the petri dish if needed.
  9. Use the beaker beside the light shelves to pour some deionized water into the flat, this will help the soil to keep moist.
  10. Cover with a plastic tray with a dome
  11. Put the tray back onto the light shelves

Note:

  • You will need to come to water your plants until we are done with the experiments
  • Arabidopsis might need to be watered twice per week, details will be given later.
