Plant phenotyping protocol

Goal: To compare phenotypes of different plants by quantitative and qualitative methods

Materials:

  • Arabidopsis thaliana plants: Wild type (WT) and PLIP3-OX
  • Camera (phone is fine)
  • Rulers

Procedure:

  1. Obtain the plants from the instruc7on team
  2. Record the plant IDs, sowing date, and today’s date; count how many days the plants have been growing
  3. Visually observe the plant phenotypes and record the following traits in your notebook for both types of plants:
    • Plant size (diameter): Big or small
    • Plant height: tall or short
    • Leaf size: big, small, medium
    • Leaf length: long or short
    • Leaf width: narrow or wide; lengthy or round
    • Petiole length: long or short or not visible
    • Flowering: yes or no
    • Color difference: what colors?
    • Other?
  4. Use your phone to take pictures of the plants
    1. Ensure you capture the visible traits of the plants
    2. Remember, the goal is to compare the phenotypes; discuss with your team in the lab the best way(s) to take the pictures
    3. A piece of black cloth will be provided for a clean background when taking pictures
  5. Use a ruler to measure the measurable traits and record the data:
    1. Use centimeter or millimeter as the unit
    2. Measure 3-4 plants and record the data

6. Take the average of the measurements of each type of plant; Show your work
